Including an artficial intron before a cdna sequence is not the same as expressing from genomic DNA. I'll tell you for a fact that the GTC epo/albumin fusion contains no introns within the coding region of the final product. All of their constructs contain the necessary upstream and downstream elements for efficient expression. As for your broad reaching generalization about cDNA constructs... I'll leave you to think what you will.

I've never done large scale protein expression, but on the small scale cDNA has always worked well for me in COS, CHO, HEK, Baclavirus (insect), and e-coli (simple proteins). Expression depends in a large part on the choice of promotor. I have only had trouble once, and that was with a large transmembrane protein.
