L is a maintenance treatment for some while for others it leads to longer term immune memory. With Direct Immune memory was achieved under certain circumstances. With proper activation, dosing, spacing and adjuvant(s) immune memory is not only possible but highly likely. DCVax-L was deliberately under powered because the immune response starts with activation from outside of the brain then built upon because of needing a controlled immune response in the brain. Uncompromised DCs in the tumor environment are the most desirable way to activate immune response because of signaling caused by DCs that are in that microenvironment that changes it to become more immune response friendly. DCVax-L does not work by getting immature DCs into the brain where they can direct the immune response from within. This is why L often times needs help from other treatments. Those treatments can activate other immune cells better and or create clearer pathways so various cell types can gain access to the brain. Some, like checkpoint inhibitors, take the brakes off of the immune response because DCs are not controlling the signaling from within the tumor like they would be with Direct. Best wishes.

Just a follow up. In the combo trial the minimum number of treatments was reduced to 3 from 5 in the Phase 3. The combo and improved activation was likely expected to make the treatment more effective. Best wishes.
