Flipper, Great explanation! My general understanding is ADCs carry high toxicity risk. They travel around the body looking for the specific target antigen and will sometimes cross react with good cells. Or if it can't find the target will biologically break down over time and the chemo drug is released somewhere. Or it could get caught in the kidney, as the kidney filters blood for toxins, and then damage the kidneys.

That is why I strongly believe DCVax is a vastly superior therapy, because of its mechanism of action. It does not bypass the body's immune system as ADCs do. DCVax leverages the natural safeguards and checks and balances of our own immune system. DCVax informs and activates immature T cells and B cells, so the immature T cells naturally pass thru the thymus and immature B cells thru the spleen. There, the body screens out and destroys any of the self-reactive immune cells (to prevent autoimmune and toxicity). Only the safe-for-self T cells and B cells can leave. Only then are they allowed to go out to multiply into the thousands and thousands.
