A lot of this stuff is informed, and not so informed, speculation. (See Simonian articles in SA.) It's not science, lacks any rigor or testing. As already noted, the guy was touting other AD drugs too. Speculators'love these "dots". It's on the internet, it must be true.
And your "Anavex have never really explained why A2-73 would work" is correct, as S1 agonism in itself is not conclusive of efficacy. (Plenty of other S1 agonists.) If 2-73 is effective, it is likely because of either a unique bonding mode or ancillary effects (other agonism, metabolites downstream, etc). The company has thrown some speculation out there on this, but emphasizes the S1 agonism.
