I know exactly what you are thinking in terms of the brain and Direct but don’t forget they can dial it down too and give it at the optimum time from off the shelf donor made product. Oh the possibilities... don’t get me started ; ). Best wishes.
Here is a research question for you to have fun with.
A reason DCvax-L might work well in GBM is because the BBB is fairly effective at preventing DCs from entering the brain even though it allows some T-cells in. Any DC vaccine bypasses this block. Thus opening an immune channel that was previously blocked.
The question with Direct is if this still holds true. The BBB is not symetrical, I have seen nothing about the ability of DCs to leave the brain. If they can freely exit, then the idea might make sense.
BTW, remember that the Direct trial proposed for brain mets uses a slow drip system to avoid inflamation.
