Exosomes from patient samples 5 µL of 4 µm aldehyde-activated latex beads (4% w/v) (Invitrogen) were incubated with 1N11-T (12.5 µg) at 4oC overnight. The beads were then washed, blocked for 1 hr with 1% BSA and mixed with 300 µL of a 1/6 dilution of patient plasma for 2 hrs at 20oC. The beads were then washed with HEPES-saline and a 1/100 dilution of mouse anti-human CD63 antibodies (Sigma #SAB4700215) for 30 min. The beads were then washed and stained for mouse Ig and PS with a 1/500 dilution of FITC-goat anti-mouse Ig (Jackson Labs #115-095-166) and a 1/100 dilution of Cy5-annexin 5 (Biovision #1013- 200) in HEPES-saline containing Ca2+ (1 mM). Samples were screened and analyzed as described above.
A recent investigation showed that expression of CD63 positively correlates with the invasiveness of ovarian cancer.[4] In cell biology, CD63 is often used as a marker for multivessicular bodies, which are enriched with CD63. [5]
