They used much more because it was a P1 trial. One of the goals of a P1 trial is to determine the maximum tolerated dose. They also were looking at the blood concentration levels so they increased the dose to see if and where the blood concentration plateaus. IIRC that was found to be about 160 mg dose.

P2 will be more oriented towards determining the minimum and maximum effective dose.

If investing was easy, everyone would be rich by now.
