pGBM=GBM is composed of two molecular sub groups and the signature is TERT, NF1, VEGFA(B) for MES and mostly ERGF amplification as a result of overexpression of the ERGFviii protein in HLA-1&2 for classical. Both are IDHwt and both have DNA methylation in varying degrees in addition to non-methylation which is a more aggressive category with a shorter mOS. For example, non- methylated MES has an mOS of about 11.5 months. MES is a rather large category and makes up anywhere between 30-48% of pGBM depending upon the literature one reads. This bodes extremely well for the trial results especially where 45% of the categorized group(N=38 of the initial group of trial patients were uncategorized) were MGMT methylated. JMHO.
