As I've understood, the Autophagy MOA was unknown to (at least most of) us until the paper just published in Cell. Before that we relied on 'just' the Calcium Homeostasis theory and the re-connection of ER to MAM MOA.

Good observation. Until now, so true.

Actually, autophagy, as a generalized waste capture and regeneration process is rather well-known. Early on, I perceived that it might be enhanced by any of the Anavex sigma-1 receptor agonists, inasmuch as they have so many diverse, positive downstream outcomes.

Several years ago, I stated that the Anavex sigma-1 receptor agonists work and have positive therapeutic outcomes because of the enhanced endoplasmic reticulum function factor. As I expected, those posts generated a good number of criticisms, claiming that there was just no “evidence” for such biochemistries and, simply, Anavex 2-73 won’t work against central nervous system diseases. (Those critics have been curiously absent the last few days, for some reason.)

Yes, early on, after reading the many Anavex studies on murines (lab rodents), and parsing out the suggested mechanisms of actions in those papers, I could see that the Anavex story won’t be confined to just Rett syndrome, Parkinson’s disease, and Alzheimer’s. The Anavex molecules simply fix so much, by so many mechanisms. Unlike everything else thrown at those (and other CNS diseases) the Anavex molecules fix bad chemistries at the very start, with gene expression (protein synthesis), or with protein folding. That gets reaction-controlling enzymes properly synthesized. Everything downstream then works normally.

This just occurred to me. A new name for the Anavex sigma-1 receptor agonists. They are “contratoxins.” Toxins, of course, poison things. In cells, they interfere and disrupt normal cell chemistries. Pathogenicity.

The Anavex molecules are exactly the opposite. They are profound contratoxins. “Contra-” means “opposite,” or “against,” When diffusing into neurons, at their start they reverse various pathogenic chemical cascades; the things that directly cause the symptoms of CNS diseases.

As I’ve claimed before, I’m certain that Anavex Life Sciences Corp has a pile of data from stacks of rodent cages from laboratories across the world, where grad students and research interns have been dosing transgenic murines with Anavex molecules for a host of diseases and conditions.

Missling’s inadvertent (never repeated) “...tip of the iceberg...” utterance probably derived from those undisclosed studies. The Anavex pipeline page hints at the matter, with several other conditions being targeted for human trials.
