I don't know to what extent it is usually the case in the clinic, but irreversible inhibition is associated with greater potency (at the expense of increased on-target side effects). That could be a factor, but as I said, it might not necessarily play out this way in the clinic.

Thanks, that's exactly what I had assumed (irreversible might have edge in potency whereas reversible might have edge in safety). I guess it's good that the drug already showed some activity, though it remains to be seen how the drug fares against the newer agents you referenced. Hopefully with it being reversible safety will be good down the road. And hopefully ARRY will be able to convince a partner of all these things. ; )
