you are wrong. gamma secretase in an enzyme that inhibits many different pathways, including notch. it is non specific and can cause severe side effects in mutiple systems if not targeted enough. Lilly's GSI was successfull at lowering CSF abeta in an early ph 2, but there was a lot of toxicity. they ended up taking a lower dose into phase 3 and failed. that same dose also showed nothing on biomarkers in ph 2.
