Bypassing the first phosphorylation step is highly consequential because in vivo performance of this step is inefficient. Thus, a nuke that already has the first phosphate group attached, as IDX184 does, can be dosed at a much level than a nuke that doesn’t, which improves safety and tolerability for a given degree of efficacy.
