Beta-secretase is an enzyme responsible for the build-up of sticky deposits, or plaques, in the brain. The formation of these amyloid plaques is thought to play a key role in the development of Alzheimer's disease.
Beta-secretase actually cleaves APP, the resultant protein is then further cleaved by gamma-secretase to form AB42 monomers, which then combine into neurotoxic oligomers and possibly-neurotoxic plaques.
This is a good target to aim for, better than the PIII Lilly/Elan gamma-secretase inhibitor which has too many side effects. However, TTHI/Elan have their PII oral ELND-005 drug which prevents AB42 oligomers from binding with neuronal cells. I think the beta-secretase inhibitor only has potential to make a serious splash if that drug fails. Neither has the ability to clear existing plaques (Elan's AAB-001) or repair damaged neurons (nobody).
