per the article severe rash is definitely an uncommon but noted side effect of riba-interferon cotherapy.
your speculation regarding augmentation of interferon-related rash from vx-950 synergism makes sense, although I wonder why other interferon-related side effects are not augmented as well if there is some boosting of interferon activity. the metabolic pathways do not converge, with PI inhibitors metabolized via cytochrome p450 and interferon via renal clearance, so some pk interaction is not likely..hence any synergy must come at target sites (although given the unclear moa of interferon i would think any touted synergy must be an empirical observation on the part of vertex rather than one that was predicted a priori)
another possibility is the obvious - that the rash is directly related to vx-950..after all, rash has been noted to occur to one degee or another in most PIs. as to why this wasn't noted previously, well i would simply answer this is the first trial of any significant size with vx-950, and that is why the bellyaching about reserving judgement on vx-950 until larger trials are complete deserves some merit..b/c all too often side effects such as this rash won't be seen in small trials, plain and simple...i don't have to remind anyone on this board how the GI side effects bit nm-283 in the rear earlier this year, and we'll just have to wait and see how vx-950 plays out
