If you look at the adverse event you have an increase in peripheral vascular disease that suggests accelerated atherosclerosis. Plaque buildup will lead to higher chance of thrombus generation when the affected vessel is damaged and result in an embolic event if the thrombus is detached from the site. Carotid stenosis is a good example (atherosclerosis of carotid arteries generating emboli causing stroke). I think most people just assume embolus can only arise from atrial fibrillation or infective endocarditis that is where the confusion comes from. I do believe ponatinib causes endothelial cell dysfunction and this can explain the finding of hypertension together with accelerated atherosclorosis. If ariad could figure out how ponatinib does that thats a blockbuster drug lol.
