other explanations for the disparity between PFS ad OS, particularly for trials in the first-line setting.

I recently came across a review discussing the relationship between PFS and OS in colorectal cancer. The correlation is quite clear for chemo, less clear for Avastin and least clear for Erbitux.

The stem-cell hypothesis I mentioned is of course pure speculation - there is no actual evidence for it that I know of.

It's frustrating to me that these studies don't ever seem to go into cause of death to try to tease out why PFS and OS are sometimes related and sometimes not.
