JAKs

One minor opinion I will add here is that JAK1 and JAK2 have a degree of redundancy between them. Therefore, if a signaling process is depending on activation of receptors upstream of JAK2, these receptors may be shared by JAK1. This redundancy may work against efficacy, to some extent, if the drug targets JAK2 specifically (or too selectively).
