there are your exact words. you make it sound like geron is teetering-----of course there likely isnt some gaping hole in technologies presenting a slam dunk in curing cancer. nonetheless, dr. okarma sounds far more confident than you and i believe he has made statements that should have greatly answered any legitimate concerns in this area.

Detection of carcinoma of uterine cervix at the very early stage is possible by the telomerase enzyme activity.
This conclusion was made by specialists of the Lomonosov Moscow State University and the N.N. Blokhin Russian Oncological Research Center, Russian Academy of Medical Sciences, after they have investigated several dozens of this tumor samples caused by papilloma viruses.
Telomerase is the enzyme that enables the cell division for unlimited number of times. Civilized cells are capable to undergo no more than 30 to 50 divisions, and they die after that. This is due to the fact that each chromosome’s ends represent recurrent sequences (telomers). With each division, the telomer length is decreases, and when it reaches the critical value, cells lose the ability for further division. However, the telomerase enzyme works in some cells, lengthening chromosome ends and allowing the cells to divide infinitely. The telomerase acts like this in stem cells and in germ line cells, in immortal cell lines cultivated in laboratories, as well as in the majority of cancer cells (80 to 90 percent). The enzyme activity could serve as a universl marker for cancer early detection, if it were not one “but”. Some types of malignant cells maintain a constant length of their telomer by means of a different mechanism, without telomerase participation. Therefore, the question whether the telomerase enzyme can be used as an “ alarm signal” should be resolves separately for each type of tumor.
The researchers used as investigation matter 28 tumor samples received from cervical carcinoma patients. All cases were caused by papilloma viruses. The major part of tumors were at early stages but some of them had metastases in lymph nodes. For reference, the researchers used non-cancerous growth cervical tumor.
The researchers discovered telomerase activity in all cancerous growth samples, even at the earliest stages. In the non-cancerous tumor sample, telomerase activity is practically absent. Consequently, cancer cells of this type provide for their “immortality” particularly with the help of telomerase. The researchers did not discover distinctions between cervical tumors from patients with metastases and without them, therefore, it will be impossible to determine the stage of disease by telomerase activity. But it is quite expedient to use the enzyme’s activity as a tumor marker to “catch” it at an early stage.

A means to an end: Telomere maintenance and bone marrow failure
A new paper from Drs. Judy Wong and Kathleen Collins (UC Berkeley) in the October 15th issue of G&D reveals a molecular mechanism underlying bone marrow failure in the X-linked human disease, dyskeratosis congenita (DC). DC is a rare, progressive bone marrow failure syndrome that is fatal early on in life. Working with cells derived from DC patients, Drs. Wong and Collins demonstrate that X-linked DC cells are unable to renew themselves because they cannot maintain sufficiently long telomeres to support proliferation. The researchers go on to show that these defects in telomere maintenance are caused by a reduction in telomerase RNA, rather than a deficiency in ribosome biogenesis, as some other groups have hypothesized. Dr. Collins is hopeful that "our studies to understand disease mechanism will aid ongoing clinical efforts to develop disease therapies."

163L IS in trials, new sites keep cropping up every month or so.
Talk to the FDA about GRNVAC1 required IND paperwork, peer review, etc. and see if you can limit some of the delay. Take a look at the "tweaking" and see if it doesn't perhaps make some sense in retrospect to have investigated down those paths. While you're chatting with the FDA, you could see what the hold up in on the cloned meat and milk approval; you don't suppose that has anything to do with the Pres and the RR, do you?
Geron is certainly transitioning from R&D to products - and that's all I've heard him say. There are some minor products now, Roche due to sell the bladder cancer diagnostic in Europe later this year.
Certainly looks like Geron Bio-Med is ramping up and expanding. University of Edinburgh collaboration on three cell types. Work progressing on all the cell lines. Progress towards the spinal cord IND, religious right notwithstanding. Diabetes work in Canada. Feeder free patent. Two lines qualified for use, one of which is a "Bush approved line."

Geron Presents Data On HIV/AIDS Drug Candidate at the XVI International AIDS Conference

MENLO PARK, Calif.--(BUSINESS WIRE)--Aug. 21, 2006--Geron Corporation (Nasdaq:GERN - News) announced today the presentation of new data supporting the development of TAT0002, a small molecule telomerase activator for the treatment of HIV/AIDS, at the XVI International AIDS Conference in Toronto.
The data, which were presented by Calvin Harley, Ph.D., Geron's chief scientific officer, and co-authored by collaborating scientists at the Geffen School of Medicine at UCLA, confirm and extend earlier findings that TAT0002 enhances the function of key immune cells from HIV/AIDS donors. The results show that TAT0002 stimulates RANTES, MIP1-alpha and MIP1-beta, molecules that attract other immune cells to assist in the anti-viral response.
As in previous functional studies, the activity of TAT0002 was blocked by co-treatment with a potent and specific telomerase inhibitor, suggesting that TAT0002 works through telomerase activation. TAT0002 was previously shown to stimulate the production of IFN-gamma -- a molecule involved in the anti-viral response of cytotoxic CD8 T-cells specific for the HIV-1 AIDS virus -- and to enhance the ability of the CD8 cells to proliferate and kill HIV-1-infected CD4 cells.
Dr. Harley also presented data showing that TAT0002 is orally bioavailable in animals. These studies, in conjunction with pilot toxicology results, suggest that therapeutic blood levels of TAT0002 can be achieved with safe, oral doses of TAT0002.
"The new studies demonstrate that our orally available telomerase activator drug broadly reactivates anti-HIV immunity in AIDS patients' lymphocytes," said Thomas Okarma, Ph.D., M.D., Geron's president and chief executive officer. "The lymphocytes are the primary mechanism for containing HIV infection early in the course of the disease. Over time, telomere loss in these cells results in the gradual decline of their anti-HIV function, leading to clinical disease progression. Our hope is to use TAT0002 to prevent this immune decline and thereby prevent HIV progression."

China touts initial success with first AIDS vaccine

BEIJING (AFX) - China said initial test results of its first AIDS vaccine showed it could protect people against the HIV virus.
None of the participants in the clinical trial's first phase showed severe adverse reactions after 180 days and some showed immunity to the HIV-1 virus 15 days after receiving the vaccine, the State Food and Drug Administration said.
'Initially, this indicates the vaccine is effective in stimulating the body's immunity,' the administration said in a statement on its website.
Researchers are still analysing the outcome of the initial trial before deciding whether further tests would be carried out, it said.
Kong Wei, the research team leader from Jilin University, told China Daily the initial results were 'truly inspiring' although he said it is still too early to claim success.
The first phase tests began in March last year in southwestern China's Guangxi region, with 49 healthy men and women aged between 18-50 participating, the newspaper said.
The Ministry of Science and Technology said another 800 volunteers, including those from high-risk groups, will be needed for the second and third phases of the trial, the report added.
However, testing to ensure the vaccine's safety and effectiveness could take years.
China started its own research into an AIDS vaccine in 2003 and has already invested over 100 mln yuan into projects for the treatment and prevention of the disease, China Daily said.

The Bill and Melinda Gates Foundation is close to reaching a landmark agreement to start an AIDS initiative in China that's worth $200 million (U.S.), the Toronto Star has learned.
While UNAIDS estimates 650,000 Chinese are said to be infected with HIV — India and South Africa alone combine for more than 10 million cases — infectious disease experts say conditions are perfect in China for the disease to spread.
The communist Chinese government for years denied the existence of AIDS within its borders and many doctors have refused to diagnose it. The government also tried to cover up an outbreak of HIV in central China that was caused by a tainted blood-selling program.
But more recently, President Hu Jintao has visited AIDS patients in hospitals and has gone on the offence against infectious diseases.
The Gates Foundation has become one of the world's most influential philanthropies. Headed by Gates, the world's richest man, and his wife, the foundation is the world's largest with assets of more than $30 billion.
This summer, billionaire Warren Buffett pledged to give much of his fortune to Gates's foundation, effectively doubling its spending power.
Carmine Bozzi, a Gates Foundation official who has led talks with the Chinese government, confirmed yesterday that the foundation and the Chinese are in final-stage talks to sign a memorandum of understanding.
"The big problem in China is IV (intravenous) drug use," Bozzi said, adding that the Gates program would largely be based in urban centres.
While Bozzi declined to share specific details about the size of the Gates' prospective investment in China, officials with other nongovernmental organizations said it would total close to $200 million over five years.
The donation would be similar in size to the Gates Foundation's project in India, where the group Avahan — which means "call to action" — is promoting condom use, heightening awareness of high-risk sexual behaviour and improving the diagnosis and treatment of sexually transmitted infections.
China's infection rate is low, at about 0.1 per cent, but the virus is spreading fast. Last year, there were 70,000 new HIV infections in China, according to UN statistics.
"What's scary about China is how quickly the disease could spread there," said Joe Amon, head of the AIDS program with the aid agency Human Rights Watch.
"If you have 1 million people infected and perhaps another 25 million people in high-risk situations — sex-trade workers, IV drug users and gay men — and no education program, how fast could it spread?"
Amon said the AIDS situation in China is comparable to some jurisdictions in Russia, where, due to drug users sharing dirty needles, infection rates have spiked to 50 per cent from 2 per cent in just two years.
Amon said that while the Gates Foundation is dealing with China's health ministry, ultimately the justice ministry will likely determine the program's success.
"They are going to control access to IV drug users and other high-risk groups."
China has also accepted a $95 million (U.S.) grant from the Global Fund to Fight AIDS, Tuberculosis and Malaria to launch a drug program that would provide free anti-retrovirals.
A year ago it allowed the Clinton Foundation, organized by former U.S. president Bill Clinton, to establish a toehold in the country. Clinton's group signed a deal with Mchem Pharma Group of Xiamen to provide low-cost chemical ingredients to companies that make generic AIDS drugs in India and Africa.

the geron hour---in the span of one hour last night i saw: cbs nightly news with harry smith substituting for bob schieffer-----about a five minute segment on embryonic stem cells and how they may help the 27 million, just in the usa, suffering from poor to no hearing; a new merck commericial further pushing the envelope on cancer vaccines----we have seen the old merck commercial regarding cancer vaccines but this was new and stressed how a cancer vaccine is probably coming; switched to pbs and we have charley rose and panel drilling some guy who opposes hesc's; after watching rose and panel for about twenty minutes they move immediately to hiv and i think the toronto aids meeting.

wow, great post esammee. now i'm probably off base here, but it seems some abstracts or scientific papers posted have suggested when geron's cancer therapies have been combined with therapies on the market, there has been a somewhat "night and day difference" to the positive, versus the current therapies without geron's stuff----is this wrong? it seems we are just talking about subjective/relative wording here, but i thought i remembered some big differences when geron's therapies were added to the mix. i hate putting numbers to the words that i'm typing, but it seems geron's therapies when added produced results in the area of 50% superior, or definitely higher in a number of cases, versus other market therapies, either solo or in combination. yeah, 100% would sure be nice. please forgive my memory/science shortcomings.

Anyhow, the big question is the validation of the science. Geron has a major foothold in the practical aspects as well as theory and if any researcher, even in China, shows what can be done specifically with ESCs, this is of major importance to Geron.
An Important Example: Some researcher shows that ESCs are indeed immune privileged in humans. Absolutely huge, major and this would mean that the islet stuff would probably go ahead full steam.
Summary: CIRM and other ESC stuff anyplace, by anyone has the potential to be huge for Geron; a success with ESCs no matter where will be the very thing Geron needs.

Running with the $150 million loan it just got from Gov. Arnold Schwarzenegger, the board overseeing California's $3 billion stem-cell institute said Wednesday it will begin soliciting its first research proposals within weeks.
The board also passed a policy to guard against bias in awarding the grants, by requiring the institute's advisory boards to disclose ties to companies or universities doing stem-cell work.
Armed with Schwarzenegger's loan, the institute plans to begin awarding grants as soon as February, a move that elicited cheers from several of the agency's board members Wednesday at their meeting.
``We now are in business,'' said Sherry Lansing, a board members and former chair of Paramount Pictures, at the institute's board meeting. ``I can't help but be excited.''
The board plans to award 70 grants totaling $151.5 million, all of it allocated for studying human embryonic stem cells, which are derived from embryos and can grow into any type of tissue.
The institute was created by California voters in 2004 primarily to conduct that kind of research, after President Bush limited federal financing in that area. But lawsuits have held up the institute's $3 billion funding for more than a year.
The idea of imposing conflict-of-interest rules on the board's advisers has been controversial. Consumer advocates and others say the advisers should not have ties to those seeking the institute's grants to insure the agency's decisions are unbiased.
But some institute officials have been reluctant to impose such limitations, fearing the advisers might be discouraged from helping the agency.
Under the rules adopted Wednesday, the experts must disclose their investments and avoid offering recommendations about businesses, universities or others with which they have a financial, personal or professional relationship.
That's similar to a policy already in place for the institute's board. But while members of the public can review the disclosures made by the board, they won't be able to see the information submitted by the advisers.
David Magnus, who directs Stanford's Center for Biomedical Ethics, said it's not essential for the experts to be held to the same rules as the institute's board, because the experts only advise, while the board makes the final grant decisions. Moreover, Magnus said he has been generally impressed by institute's effort to avoid conflicts of interest.
Some consumer advocates aren't satisfied.
``It amounts to self-regulating,'' said Jesse Reynolds of the Center for Genetics and Society in Oakland.
State Sen. Deborah Ortiz, D-Sacramento, backed a constitutional amendment last year that would have required the board members to divest their investments in stem-cell companies. But Ortiz later dropped the requirement.

thanks Paula, yeah the button isnt showing up at ihub or silicon but that is probably because i'm not logged in over there. the main thing is i'm the only one having problems at siliconinvestor.com and there are no problems here. everyone can post. everyone can start a new account. both places. i'm happy. i feel for dave.

I don't doubt the errors are happening, but we have hundreds of registrations a week. Let me work on this and we'll get it solved!

esammee, i did what you said but couldnt find the "mail button. maybe that is because "the page cannot be displayed" is coming up both at ihub and silicon, but not a yahoo so i dont think i have a computer problem. silicon and ihub may have received the new equipment and are installing as we speak. the good part is folks can still post at both places and start new accounts at both places. i'll get with Paula asap, kind of a wild day around here------but i did receive another email from dave and it looks like we are just about there------also, when we can send free messages a week from friday i will send all communications between dave and i-----it is interesting-----i'll give you my email address also and you can give it to anyone you see fit. (that's a pretty cool feature at ihub-----we can talk in private without exchanging email addresses)

Enzyme has power to 'save and kill'

A special Jekyll-and-Hyde-like enzyme found in the body holds the key to long life but also has the power to kill, a conference will be told.
The enzyme - a so-called molecular fossil known as telomerase - was discovered during the 1980s by former Melbourne researcher Professor Elizabeth Blackburn.
It was known to be overactive in more than 80 per cent of cancers.
But Prof Blackburn, now at the University of California, will tell the International Congress of Human Genetics in Brisbane that the enzyme is also linked to heart disease.
Telomerase levels control the fine balance between ageing and immortality, with a controlled balance essential for cells to replace themselves.
"Without telomerase, chromosome ends dwindle away as cells divide, and this shortening of chromosomes contributes to the wear and tear of age," Prof Blackburn said.
She said recent clinical and experimental observations suggest a link between the enzyme and diseases of ageing like heart disease.
"If we could reactivate telomerase function in ageing cells, we may be able to slow down the ageing process by repairing damaged tissues and organs," she said.
But telomerase is the Dr Jekyll and Mr Hyde of cells.
It can help us live longer, but too much of it in the wrong cells is associated with cancer, Prof Blackburn said.
She said developing new drugs that interfere with telomerase in tumours may be an effective way of fighting cancer.
The scientist is expected to unveil details of her research program at the conference in Brisbane next week.

DR. OKARMA: Right. Well, the, the application here for our own IND is what's occurring. The steps first are to submit to the *RAC - that has happened. Once the RAC buys off of the application, then we file with the agency, and that is on track to happen very soon. So we would hope to actually begin enrolling subjects by end of the year under our own IND. We have not yet announced the actual tumor type; I can tell you that it is within the hematologic malignancy arena and the reason for that is the trial is designed to demonstrate a direct impact on tumor progression, which, as you know, has really not been described by anyone in the cancer vaccine space before and that is very difficult to do in prostate cancer because it's hard to get objective measures of tumor progression. So that's as much as I can tell you now, and when we're actually enrolling, we'll announce it.

wow, thanks for the kind words. this downtrend is done i hope as buying the rumor might start real early here--------the potential/probable universality of therapies getting after these awesome targets is far beyond anything ever accomplished in cancer medicine. yeah, the science folks on this board are too much-----really great.
