Probably oversimplified.

Trial size calculations are just a statistics problem.

Maybe closer to truth for a P3 trial where enough responder data is available to assess effect size before designing a P3 trial.

Without a good sense for effect size, then the trend would be towards a larger trial not to flunk a first attempt.

Anavex clearly must have felt they knew enough about expected effect size to go for the smaller trials.
