He may have a very good reason to sell, but it's a terrible PR problem for Missling.

He has to be able to the question from institutional investors: why should we invest in AVXL when one of your directors exercised his options and sold the majority of his holdings?

Related question: if you all are so confident in what you are doing why would an insider sell and potentially leave so much money on the table?
