$ONCX stopped reporting in 2018. I had small communications with the CEO a couple years ago but he acted like he could give a $hit. Now the email is dead. Bright side, as of 2018 we had patents and hard hours in research/studies that are some killer assets. CEO was a crook and a dipstick, convertible debt brought it 2 its knees and has been here since.

Notice how prolactin hormone increases proliferation from baseline but Prolanta drug decreases proliferation from baseline. Also, on the far right, notice how the Prolanta drug fairly effectively cancels out the prolactin hormone when both are present. Like I said, Prolanta is a cool molecule with some intriguing biological effects in animal studies.

Abstract
Abnormal activity of human prolactin (PRL) and its membrane-associated receptor (PRLR) contributes to the progression of uterine carcinoma. However, the underlying mechanisms are not well understood, and current means of targeting the PRL/PRLR axis in uterine cancer are limited. Our integrated analyses using The Cancer Genome Atlas and Genotype-Tissue Expression (GTEx) databases demonstrated that a short form of PRLR (PRLR_SF) is the isoform predominantly expressed in human uterine cancers; expression of this PRLR_SF was elevated in uterine cancers in comparison with cancer-free uterine tissues. We hypothesized that the overexpression of PRLR_SF in uterine cancer cells contributes, in part, to the oncogenic activity of the PRL/PRLR axis. Next, we employed G129R, an antagonist of human PRL, to block the PRL/PRLR axis in both PTENwt and PTENmut orthotopic mouse models of uterine cancer. In comparison with control groups, treatment with G129R as monotherapy or in combination with paclitaxel resulted in a significant reduction of growth and progression of orthotopic uterine tumors. Results from protein profiling of uterine cancer cells and in vivo tumors revealed a set of new downstream targets for G129R. Our results showed that G129R induced sub-G0 population arrest, decreased nascent protein synthesis, and initiated FOXO3a/EIF-4EBP1–mediated cell death in both PTENwt and PTENmut uterine cancer cells. Collectively, our results show a unique pattern of PRLR_SF expression predominantly in uterine cancer. Moreover, FOXO3a and EIF-4EBP1 are important mediators of cell death following G129R treatment in uterine cancer models.

The Company is halting enrollment in its Phase I clinical trial of Prolanta for the treatment of ovarian cancer as a result of injection site reactions at the current dose level. No other adverse effects or signs of toxicity have been observed for the drug.

Two subjects enrolled in the second dosing group, 1 mg per kg of body weight, have experienced inflammation and edema at the injection sites that resulted in their withdrawal from the study. Prolanta is injected subcutaneously daily during the treatment period. The Company believes that a reformulation or alternative dosing method of the drug will be required to continue the clinical evaluation of the drug, and such changes may require additional preclinical toxicity studies and additional clearance by the U.S. Food and Drug Administration. The Company does not have the financial resources at this time to conduct such additional studies.

That's it. Two people in one of the dosing groups got a rash at the subcutaneous injection site. This was the straw that broke the camel's back for a company in dire shape. What should have been a slight, temporary setback resulted in the entire trial being permanently shut down. No question about it- it was definitely more of a failure of management than it was a failure of the drug.

Good Lord no. "Prolanta" (PRL-inh G129R) is a cool molecule with some intriguing biological effects in animal studies. That is approximately one quadrillion miles away from an FDA-approved product. This paper is from an academic research team in Mexico City using donated material. Oncolix is defunct. As far as I know, no companies are currently developing Prolanta for any indication. As far as I know, ONCX does not have any employees, officers, or BOD, and they are way overdue with SEC.

There are currently no cures or effective treatments for lupus, except for long-term steroids, which then results in a whole slew of steroid-induced complications.

This is an overstatement by me, which is probably the result of personal bias from my experience with a few tough cases. In fact, there has been moderate success using hydroxychloroquine for some cases of lupus. Methotrexate and similar drugs can help to avoid flares without using frequent steroids- but at their own costs. Rituxan and other non-steroid immunomodulators have an important role. It's the severe cases of lupus with frequent flares of debilitating arthritis and/or kidney dysfunction where options are extremely limited beyond aggressive steroids. There is definitely a need for more effective therapies with better side effect profiles.

2.3. Prolactin Hormone and Inhibitors
We used murine recombinant PRL (National Hormone and Peptide Program, NIH).
PRL receptor activation was ablated with an inhibitor (PRL-inh, G129R), a recombinant
analog of the PRL with a single amino acid substitution to create an antagonist of the
PRL receptor, which was donated by the manufacturer (Oncolix, Houston, Tex, USA) [22].
Stattic (Cell Signaling Technology, Danvers, MA, USA) is a nonpeptidic selective STAT3
inhibitor. This small molecule prevents the binding of tyrosine-phosphorylated peptide
motifs to the STAT3 SH2 domain [23].

Long story short, prolactin receptor is expressed in all B cells, but its expression is increased in the type of B cells that are implicated in the development of lupus. Prolactin binding to these receptors seems to rescue those cells from the suicide pathway, thus increasing lupus-type reactions. If "Prolanta" is used to inhibit binding of prolactin with its receptor, then potential lupus cells proceed through the cell suicide cycle.

These are just mouse cell studies, but the suggestion is that prolactin inhibitors like Prolanta could slow or prevent progression of lupus. Lupus is an auto-immune condition that affects millions of people world-wide, mostly women. It can be highly debilitating and is a frequent cause of kidney failure. There are currently no cures or effective treatments for lupus, except for long-term steroids, which then results in a whole slew of steroid-induced complications.

We found that all B cell maturation stages
in bone marrow express the prolactin receptor long isoform, in both wild-type and MRL/lpr mice,
but its expression increased only in the immature B cells of the latter, particularly at the onset of lupus.
In these cells, activation of the prolactin receptor promoted STAT3 phosphorylation and upregulation
of the antiapoptotic Bcl2a1a, Bcl2l2, and Birc5 genes. STAT3 binding to the promoter region of these
genes was confirmed through chromatin immunoprecipitation. Furthermore, inhibitors of prolactin
signaling and STAT3 activation abolished the prolactin rescue of self-engaged MRL/lpr immature B
cells. These results support a mechanism in which prolactin participates in the emergence of lupus
through the rescue of self-reactive immature B cell clones from central tolerance clonal deletion
through the activation of STAT3 and transcriptional regulation of a complex network of genes related
to apoptosis resistance.

This transferred technology was modified by our contract manufacturer, which may reduce any royalty related to the use of the bacterial expression system.
