Philip E. Thorpe: 1951 – 2013

Peregrine Pharmaceuticals today mourns the loss of Dr. Philip E. Thorpe, scientific advisor to the company and inventor of our phosphatidylserine (PS)-targeting technology on which our lead drug candidate bavituximab is based. Phil was a Professor of Pharmacology at the University of Texas Southwestern Medical Center, was associated with the Harold C. Simmons Comprehensive Cancer Center and held The Serena S. Simmons Distinguished Chair in Cancer Immunopharmacology. Phil’s achievements were numerous and included advances in drug targeting, angiogenesis, and antibody-based therapeutics which had a global impact. His loss will be felt deeply by all of us.

Phil’s research focused on the development of novel drugs targeting tumor blood vessels. It was his laboratory that made the remarkable discovery that a fatty lipid molecule, phosphatidylserine, is preferentially expressed on cancer blood vessels, where it can serve as a target to increase the specificity of drugs to the tumor.

Phil was firmly dedicated to the translation of his novel concepts in drug design into practical drug therapies for cancer, imaging agents, and anti-virals. His expertise covered a wide range of fields including protein engineering, synthetic chemistry, pharmacological testing, cell biology, and immunology. Five drugs developed wholly or partially in Phil’s laboratory have entered clinical trials and he is included in over 200 issued and pending worldwide patents, including dozens in the U.S. He is the author of more than 200 publications including articles in the fields of drug targeting, angiogenesis, and antibody-based therapeutics.

Phil was a tireless advocate as a scientific advisor to Peregrine for over 15 years.

Among other honors, Phil was one of the first recipients of the Pierce Immunotoxin Award, presented every two years for outstanding contributions to immunotoxin research, in 1998, and he received the Texas State Legislature Award for Research Excellence in 1997 as well as the American Cancer Society Award of Excellence in 1999.

Phil graduated summa cum laude with a Bachelor of Science in Pharmacology from the University of Liverpool in 1972, and he received a Ph.D. in Immunology from the Clinical Research Centre in London in 1976. He served as a Medical Research Council Fellow at Chester Beatty Research Institute in London (now The Institute of Cancer Research) until 1981, then as Director of the Drug Targeting Laboratory, Imperial Cancer Research Fund in London until 1991, when he first joined UT Southwestern as Professor of Pharmacology. Phil also served as Associate Director of the Center for Molecular Medicine at Maine Medical Center Research Institute from 1998-1999.

Phil will be sorely missed as a colleague and friend. At Peregrine, each of us is pursuing Phil's dream of bringing important drugs to patients who need them based on his basic research. We are more motivated than ever to see this dream through.

TUSTIN, 8/14/13: Peregrine Pharmaceuticals, Inc. (NASDAQ: PPHM) today announced the presentation of preclinical data and ongoing drug development activities highlighting the immune-stimulatory mechanism of action of phosphatidylserine (PS)-targeting antibodies, such as the company's lead drug candidate bavituximab. Data from this presentation showed that PS-targeting antibodies block a primary upstream immune checkpoint, thus inhibiting PS from engaging its natural receptors on immune cells, resulting in multiple downstream immunostimulatory effects. Bavituximab is the company's lead PS-targeting investigational product and is currently being evaluated in several solid tumor indications with the initiation of a Phase III trial in second-line non-small cell lung cancer (NSCLC) anticipated by calendar year-end. This presentation was made today at Cambridge Healthtech Institute's Immunotherapies Congress in Boston, Massachusetts.

The presentation titled: "Engagement of Phosphatidylserine (PS) by PS-targeting Antibodies Blocks an Upstream Immunosuppressive Checkpoint in the Tumor Microenvironment; Inducing Multiple Downstream Anti-tumor Response Mechanisms" detailed the role of exposed PS as an important upstream immune checkpoint. Administration of PS-targeting antibodies was shown to mediate pro-inflammatory cellular and cytokine responses that convert immunosuppressive cells known as myeloid derived suppressor cells (MDSCs) into tumor fighting (M1) macrophages. PS-targeting antibodies also facilitated the maturation of dendritic cells capable of presenting tumor antigens to T-cells to facilitate tumor-specific cytotoxicity. These multiple anti-tumor effects were further shown to occur without the side-effects associated with systemic immune activation. This was presented by Jeff T. Hutchins, Ph.D., VP, Pre-Clinical Research of Peregrine Pharmaceuticals.

"We are pleased to be presenting this compelling data to investigators of the oncology and immunology community as these results clearly demonstrate the immune-stimulatory effects of bavituximab," said Dr. Hutchins. "These data have shown the breadth of bavituximab's potential benefits as an immunotherapy and have led to the implementation of a supplemental clinical development strategy that explores the synergies between bavituximab and other immunotherapeutic drugs. The first part of this plan began in June with the initiation of several proof-of-concept preclinical studies constructed to read out in the next couple of months in order to assist us in designing a clinical trial examining a combination of bavituximab with an approved immunotherapy. In addition, we are leveraging data from our ongoing investigator-sponsored clinical trials to garner additional insight to support our development activities. We believe that this immune-stimulatory data shows that bavituximab has the potential to be combined with multiple downstream immunotherapies, such as Anti-PD-1, Anti-PDL-1, Anti-CTLA-4, IL-2, or PAP-GMCSF, to provide increased therapeutic benefit."

In addition, Dr. Hutchins reviewed the supporting rationale for the combination use of bavituximab and the chemotherapeutic agent docetaxel. Citing the synergistic immunotherapeutic mechanisms of each of bavituximab and docetaxel, the presentation summarized the promising data generated to date that further validate this combination therapy and provide additional scientific rationale that supports advancing this program into Phase III.

"These data clarify our understanding of bavituximab, put recent clinical results into greater perspective and allow us to supplement our current drug development strategy in order to enhance bavituximab's therapeutic potential," said Joseph Shan, VP, Clinical & Regulatory Affairs of Peregrine. The hallmark of apoptosis, phosphatidylserine (PS) exposure promotes non-inflammatory phagocytosis of dying native cells. This signaling pathway appears to be exploited by tumors resulting in the suppression of both innate and adaptive tumor immune responses. Administration of PS-targeting antibodies mediates pro-inflammatory cellular and cytokine responses that reverse this immunosuppressive checkpoint without the side-effects associated with systemic immune activation. Bavituximab is a late-stage clinical PS-targeting antibody that has demonstrated promising signs of anti-tumor activity and extended survival in patients with NSCLC and breast cancer.

SK 8-13-13: “We have very active partnering discussions around our 2 lead programs. Our stated goal is to ideally have a partner for both programs, Cotara, and really with all the activities going on around Bavituximab, it’s going to be necessary. We’re in a position to run a Phase III for Bavituximab and to run that study by ourselves if a partner doesn’t come on board or if the right terms aren’t on the table. Having said that, our stated goal is to identify primarily ex-U.S. partners; we’d like to at least maintain co-promotional rights here in the U.S. and there’s some significant upside at the end of the development road here. So, you know, they all have their own tone & tenor, and we’re very actively working toward it.”

I know its not over till the fat lady sings but she has an appointment and the mic will be on for all in attendance to hear. Dr Jedd Wolchok does not take such interest in the MOA of any drug but the question is the MOA of Bavituximab may be unlike any other drug that Dr Wolchok has ever seen before....

...just listen to the late Dr Thorpe for some early insights and let the immune system get back to work
