As compared to the first-generation HCV PI’s, TMC435 is more specific to the HCV protease and hence is less apt to cause the kind of off-target effect on a host protease that jellybean is talking about. Still, I don’t think we can entirely rule out the possibility that some nuke+PI combinations could fail due to the PI interfering with the metabolism of the nuke’s prodrug.
