The placebo affect will be stronger for those with less severe Rett syndrome symptoms than those girls with more severe symptoms,

I'm not suggesting the trial result depended on the RSBQ placebo effect. It straight failed on CGI.

There may be a ceiling effect on the dosed arm that would not be present in the placebo arm. No matter what, a high placebo effect is never good news for stat sig results.
