But most knockout mice aren’t actually ‘absent’ of the target gene. The gene is inactivated, mutated, or otherwise altered. In knockout mice, there is often still an effect when presented with its respective agonist. The effect is simply muted or not effective enough to return the gene’s function.

Knock out may mean loss if Sigma-1 functionality. That doesn't say where in the biologic process that loss occurs. It may mean the gene expression is suppressed not the capability is eliminated. If that is the case 2-73 has been shown to increase the expression of the Sigma-1 gene. Perhaps that was sufficient to overcome what ever was suppressing the gene expression.

You argument is based on insufficient information.
