From this alone, I can tell you do not understand what a non linear response is.

Specially a U shaped response.

AVXL will need another large P3 trial - 1000 patients where they have to use a single dose that they want to recommend for approval and get concrete results to that the dose and drug works.

A P3 Precision Medicine trial as I described could actually be quite small on the assumption patient selection criterion for response are reliable and effective. Probably n < 300.
