Yes, that's the current research thinking.

In addition: while donepezil (itself, not its metabolite) has a relatively strong S1 binding affinity, its S1 selectivity is deemed to be relatively poor.

These are some other reasons why its properties as an S1 ligand are considered somewhat incidental. And why it wouldn't necessarily interfere with other S1 ligands.
