Regarding the first bit in bold, I have been wondering why the endogenous SIGMAR1 system ends up needing a notch from something like A2-73.

Nice to have an explanation for that.

Still though there are many theories as to what might be the original cause or causes for the inflammation/stress occurring. But will be a good result if A2-73 gets proven to lend a helping hand in restoring balance and stop further decline.

Thanks for the summary my friend. While some of this stuff is well over my head, I take it you are excited about what Dr. Hampel revealed @ AAIC last week? And having this data in-hand and able do analysis, has it changed your future outlook on Anavex and chance of trial success?

I have finally have gotten a chance to take a breather and on a mandatory 4 days R&R and able to catch up on my investments. They have a taken a backseat to firefighting> typical summer activity's for me....sleep,eat and firefight,shower once a week....beer drinking mixed in here and there. Lucky for me, I'm sitting outside a hotel in Redding,Ca with a 12 pack of Pabst Blue Ribbon....its 110 in the valley right now so they are going down smoothly. Tough to keep cold though, so gotta drink quick. Have a feeling I wont get the full 4 days though.....I can see the smoke column from the fire we have been on the last 9 days (about 60 miles from where I am right now), and it not looking too good up there. Typical.

Key element of the bio environment that confirms the MoA:
• Under chronic cellular pathological stress the endogenous ligands which agonize the Sigma deplete.
• Anavex 2-73 acts to replace the depleted endogenous ligands

Well worth taking the extra 6 months to confirm the MoA and genetic condition before starting the larger trial, in order to know what to expect going in. Much better than data mining after the fact!
