Just wondering could an individual's dosing variability be related to feeding and by their stomach acidity?



Bioavailable absorption of Anavex 2-73 in the walls of the gastrointestinal tract (stomach, intestines) is probably only slightly influenced or controlled by pH levels. And GI microbes may moderately control acid/base levels in various lengths of the GI tract. But those would be variable, from various meals being digested and various microbes thriving or being suppressed by the food.

Almost surely absorption is not a factor of stomach acidity, inasmuch as very little is absorbed in the stomach; only lower, primarily in the small intestine, which is not so acidic.

The variables controlling Anavex 2-73 absorption into the circulatory system need to be discerned and understood.

But just as important, if not more so, is diffusion or transport across the blood/brain barrier, and then into neurons themselves. Concentrations in the blood may not be exactly controlling. The only Anavex 2-73 molecules that count are the ones that get inside neurons, where they work their chemical magic.

The issue of diffusion and transport across the plasma membrane into cells is a complex topic under continual investigation. Many, if not the majority of drugs, must effectively get inside cells to work. Cells have a number of structural and mechanistic arrangements to prevent chemical incursions. Those can be genetic, with some people having cell chemistries that either favorably or unfavorably keep alien molecules and ions out of cells.

But, there may well be bacteria or fungi in the gut that facilitate drug absorption, perhaps by the excretion of particular proteins that bind to the drug, making it easier to absorb.

It’s certainly not just a gut pH factor.

PA, I think in the end we will find that 2-73 on its own is the solution with no help from any of the other faux AD drugs.
