It's not clear to me what the HDAC path is that leads to AD improvement.

But then, as I understand it, it's not clear to anyone what the exact path is that leads to AD in the first place. Some of the consequences are clear, the Tau tangles and the buildup of Aß. How those things happen is still not known.

The 2-73 theory is that due to the ER mitochondrial disconnection proteins are being misfolded causing the proteins that clear out Aß to be less effective and resulting in excess Aß buildup.

How close that theory is to the true mechanism of AD is not clear to me.
