This next cohort is a break from the escalation they have been following up to this moment. Someone correct me if I'm wrong but I believe previous cohorts have ranged from 35% to 50% increases, while this is 67%. Surely previous cohorts did not show grade 2 AEs, yet no cohort increased more than 50%. Maybe the first cohort with its problem set the parameters for the next 8 cohorts, and only now are they confident enough to escalate according to the original protocol guidelines.
