Activation of the sigma-1 receptor ameliorates neuronal ferroptosis via IRE1a after spinal cord injury

Highlights

• Our study found that Sig-1R activation effectively promoted the recovery of motor function in mice after spinal cord injury, attenuated neuronal apoptosis, reduced the production of pro-inflammatory cytokines and iron accumulation, and inhibited ferroptosis in spinal cord tissues after SCI in mice.

• Ferroptosis and IRE1a were significantly upregulated after spinal cord injury, and sigma-1 receptor agonists were able to ameliorate this result through the elimination of inositol-requiring enzyme-1 alpha (IRE1a)-mediated neuronal ferroptosis.

• Therefore, sigma-1 receptor activation could attenuate ferroptosis after spinal cord injury by reducing IRE1a and improve functional recovery after SCI, providing a new therapeutic strategy for the treatment of SCI.
