The Story is Anavex Molecules, NOT Sigma-1 Protein

The more senior researchers/biochemists told him that Sigma had already been explored in the 1970's and there was no medicinal potential in this receptor site, that he was wasting his time.

And, of course, that was the case back when the sigma-1 receptor protein was discovered. It was "just there." Of no known function.

Of course, that's not the case today. The story is not this particular protein; it's Anavex's proprietary ligands that attach to the protein and induce all sorts of favorable 'downstream' processes.

Wonderfully, unlike most drugs the Anavex molecules easily cross the blood brain barrier and also easily diffuse into the cytosol, easily crossing the plasma membrane. They get themselves to the cellular venues where they can act. And, all the while failing to produce adverse events in the organism; no side effects of any consequence.
