you think Pre-084 is an antagonist? I guess > 100 papers about the drug are wrong. Thank you for setting the record straight. Pre-084 is neuro-protective, helps cognition/learning, induces autophagy, etc. --- all things we want A273 to do. So if you are right and PRE-084 is an antagonist, instead of the agonist scientists believe it to be, then the entire MOA of Anavex2-73 is wrong.

The paper results likely have no bearing on Anavex's chance in AD. A proper way to think about the new paper is to challenge existing belief systems. IF S1R agonism is good THEN increased production of Amyloid beta is disconnected from the positive effects on cognition/learning. Could it be that the amyloid deposition does not have a negative effect on cognition. Therefore amyloid-PET as a biomarker would not correlate much with AD progression. Perhaps, A273 will not clear amyloid but still lead to profound slowing of cognitive deterioration (via other cellular pathways involving S1R agonism or muscarinic effects). I think my AD patients would rather have stable ADAS-Cog scores and mildly more amyloid in my head then clear the amyloid and progress more rapidly.
