Sequestration refers to witnesses not being allowed to be present in the courtroom whilst other witnesses are giving testimony (or at all) except during THEIR testimony. They are also nott allowed to read rough transcripts of the court reporter (overnights are called 'roughs') so they cannot (or are not supposed to) learn what other witnesses testified to at trial.
This supposedly prevents them from tailoring their testimony in view of what others have testified to before them. It is of very limited effect, IMO and IME. For reasons I won't go into at this point, butt which ought to be self-evident.
