This is an accurate statement. The exact, "downstream" reaction cascades that facilitate restoration of normalized homeostasis are not yet fully characterized.

Nonetheless, the mechanisms by which Anavex sigma-1 receptor agonists initiate these processes are understood. As the authors note, "...it modulates Ca2+ exchange between the ER and mitochondria by interacting with inositol-1,4,5 trisphosphate receptors (IP3Rs)." Mitochondria and endoplasmic reticula are physically and chemically re-connected allowing calcium ion exchange, a crucial signaling and reaction-supporting mechanism.

Knowledgeable biochemists have not questioned the Anavex MOA in any way. Anavex sigma-1 receptor agonists do restore normalized neuron homeostasis in diseased neurons. When biologists, as with those with Anavex, make claims of homeostatic restoration, the very first question other biologists will ask, "OK, how so? Really? That's pretty remarkable Tell the mechanism of action, if you will. How could that be?"

And, the Anavex PhDs have done just that --- without a single posting or paper questioning the described MOA in any way. Solid science, up, down, and side ways.
