IMHO, it is a possibility, as it only affects the analysis.

"[2.] Would they also need or want to increase the trial size?"

IMHO, I don't think so, because it would be a major thing to change protocol that affects the ongoing trial proper at this point (like trial size). Besides, I think they likely already have plenty of patients to make determinations with the new immune response indicators.
