HCV-796 Review and Analysis
by: upndown1313
Long-Term Sentiment: Strong Buy 11/12/05 11:59 am
Msg: 69666 of 69733
First of all, just lt me say I have over 20 years experience in pharmaceutical research, development and supply. For my current company, which I won't identify, I do technical analysis of new drugs and technologies from feasibility, cost and risk assessment. I know very few people who have my backgound and expertise. My company is not involved with VPHM so there is no conflict of interest. All my DD is based on what has been in the public domain and my own experience.
The HCV-796 results are very exciting. My first take is that they have much work to do to improve the absorption phamacokinetics of the compound. The compound appears to be dissolution rated imited from the simple capsule dosage form since it is very water insoluble (a bad thing) but has high biological membrane permeability (a good thing). I know this because DeRosen said it during the CC and and also indicated that there was still a significant fed/fasted effect on absorption. They might need to partner with special drug delivery company like Elan to optimize the formulation for commericial use. Optimizing the formulation would result in increasing systemic expsoure, reducing patient variablity and decreasing HCV viral load and potentially improving efficacy. It is not yet known how viral load reduction and efficacy are related.
They will not need to optimize the formulation to continue the upcoming Phase IB studies with PEG-interferon. It is in their best interest to get a better formulation into Phase II, however, this won't be a requirement. It could be that the 2 patients that had greater than 2log10 reduction (> 99%) in viral titers were jsut good absorbers of the drug and had greater systemic exposure, but that is speculation on my part w/o seeing the data. I wish an analsyt would have asked that question.
The really good news is that this is the first non-nucleoside polymerase inhibitor that has demonstrated significant viral load reduction in man and was well tolerated with no dose limiting toxicity. It should be pointed out , that the safety leg of the Phase IB trial remains blinded, so we don't know why 3 of the patients dropped out of the study but toxicity is a concern for all compounds, even after FDA approval.
With respect to potency as measured by viral load reduction in man we can rank the current HCV inhibitors as:
Vertex (protease inhibitor)
> VPHM (non-nucleosise polymerase inhibitor)
> Idenix (nucleoside polymerase inhibitor
> PEG-Interferon (SGP and Roche)
Combinations of the above xave been shown to have better results (Idenix and PEG-Interferon results just released). Side effects w/ interferon are substantial (Just ask Irc) and also noted with the Vertex compound. VPHM's HCV-796 appears to be beeter tolerated based on the preliminary information. Therefore, I speculate that combinations of HCV-796 with VTX-950 at a lower dose would be preferred (very preliminary assessment)since VTX is more potent, more toxic and has difference mechanism of action. Immediate discussions here of this potential partership are premature at this time.
Overall, we need to wait for more results, but the preliminary results are highly encouraging. I expect the compound to move to Phase II and I expect the results with PEG-interferon to be even better. It would be great to see protease-polymerase inhibitor combinations ASAP, but I don't know if that will be possible from business perspective. Certainly that would be in the best interest of HCV patients.
With respect to stock price, both Idenix and Vertex shares are doing quite well based on HCV potential alone and no significant pipeline earnings currently. VPHM should eventually be priced >$40 when considering Vancocin earnings and HCV-796 potential in 2006. At this point, I think that the risk of HCV-796 not moving to Phase II is fairly low.
The fundamentals of VPHM have improved dramatically t

CNBC just said n-gas prices up due to cold weather hopefully just the start of the cold and the price of n-gas.n-gas stocks look good today
Head Higher since I just rebought on fri. after selling at 19.50 I think I missed the recent lows...Earnings come out soon and they should look good..Recent upgrade from somebody forgot who right now..been watching it trade and its looks strong too strong for I wanted it lower... just threw in the towel since it refused to go lower..IMHO yes it heads higher
According to the weather chanel there is going to be a cold snap coming down from the north this weekend and into next week..Maybe people will start turning on their heaters this may depleat the ngas reserves a little..Do you think todays report was false from the goverment to lower the price of gas before the winter season?? Three weeks ago everybody loved ngas stocks now it seems everybody hates them and doom and gloom over the sector is coming as another poster said there is blood in the streets.
Would everyboday please turn on their heaters and start useing some ngas...ug ugly on my side bought some EPEX now my largest holding and just now rebought some VPHM and took a small bite on EGY also bought TGE past few days..

When this board was started I had made the last payment on my house with profits from the stockmarket (post # 9)..I think it is only a matter of time before as people have posted on this board, that the houseing market goes down..Today is still a sellers market not a buyers so what I have started to do is move cash from stocks into secure bank CDs I hope we have another boost in OIL and GAS stocks in the next few months as old man winter sets in , at that time I plan to get out of stocks and go into cash and in a year when the houseing market pops buy another house when it is a buyers market and when they are hurting for sellers..I live in Calif which is so overpriced I cannot tell you because if you dont live here you would not believe it...I look out my window and accross the street (75 yards) the owner is selling his house for 1.1 million ( I still cant believe the price) will it sell I dont know? Probably.
I would like to buy another house in oregon still looking around for in 1 year Calif will still be overpriced and Oregon may be a good bet.
thanks doubloon for the update holding and hopeing for the best to come
