Researchers at the University at Buffalo say they have identified the single transcription factor or “master switch” that initiates the critical myelination process in the brain. Their study (“Transcription factor induction of human oligodendrocyte progenitor fate and differentiation”) is published online in the Proceedings of the National Academy of Sciences (PNAS).
The identification of this factor, SOX10, in human brain cells brings scientists closer to the goal of treating multiple sclerosis (MS) by transplanting into patients the brain cells that make myelin, according to one of the authors of the paper.
“Now that we have identified SOX10 as an initiator of myelination, we can work on developing a viral or pharmaceutical approach to inducing it in MS patients,” said Fraser Sim, Ph.D., senior author on the paper and assistant professor in the UB department of pharmacology and toxicology in the School of Medicine and Biomedical Sciences. “If we could create a small molecule drug that would switch on SOX10, that would be therapeutically important.”
Stem cell therapy is seen as having dramatic potential for treating MS, but there are key obstacles, especially the length of time it takes for progenitor cells to turn into oligodendrocytes, the brain’s myelin-making cells. Using currently available methods, added Dr. Sim, it can take as long as a year to generate a sufficient number of human oligodendrocyte cells to treat a single MS patient.
That’s partly because there are so many steps: the skin or blood cell must be turned into induced pluripotent stem cells, which can differentiate into any other type of cell and from which neural progenitor cells can be produced. Those progenitor cells then must undergo differentiation to oligodendrocyte progenitors that are capable of ultimately producing the oligodendrocytes.
“Ideally, we’d like to get directly to oligodendrocyte progenitors,” continued Dr. Sim. “The new results are a stepping stone to the overall goal of being able to take a patient’s skin cells or blood cells and create from them oligodendrocyte progenitors.”
Using fetal (not embryonic) brain stem cells, the UB researchers searched for transcription factors that are absent in neural progenitor cells and switched on in oligodendrocyte progenitor cells. While neural progenitor cells are capable of producing myelin, they do so very poorly and can cause undesirable outcomes in patients, so the only candidate for transplantation is the oligodendrocyte progenitor.
“The ideal cell to transplant is the oligodendrocyte progenitor cell,” said Dr. Sim. “The question was, could we use one of these transcription factors to turn the neural progenitor cell into an oligodendrocyte progenitor cell?”
To find out, they looked at different characteristics, such as mRNA expression, protein and whole gene expression, and functional studies. The team narrowed it down to a short list of 10 transcription factors that were made exclusively by oligodendrocyte progenitor cells.
“We describe, to our knowledge, the first comprehensive study of transcription factor expression and function by purified neural and oligodendrocyte progenitors obtained directly from human brain tissue,” wrote the investigators. “We have identified those transcription factors capable of regulating oligodendrocyte progenitor fate and establish that among these, only SOX10 was capable of comprehensively inducing oligodendrocyte fate both in vitro and following transplantation into a model of human leukodystrophy. Thus, viral and pharmacologic approaches to increasing SOX10 expression likely will improve the outcome of human transplant therapy.”
“In MS, first the immune system attacks the brain, but the brain is unable to repair itself effectively,” noted Dr. Sim. “If we could boost the regeneration step by facilitating formation of oligodendrocytes from progenitor cells, then we might be able to keep patients in the relapsing remitting stage of MS, a far less burdensome stage of disease than the later, progressive stage.”

Two points. Number one is that I'll bet this really cuts down on complaints about the food, people mistreating the waitresses and bad tipping. And number two, I'd be willing to bet that this place will NEVER be robbed.
JLHammer, That would really be great. It is just mind boggling, the many uses of this technology that can combat and possibly defeat such terrible diseases. So many, have spent their entire lives, trying to find a cure for these horrible conditions that effect so many people, all over the world. I might be wrong, but I have a feeling that something very big is getting ready to happen for our little fledgling company. And it just might change the world in a really good way. I am proud to be a teeny, tiny, little part of that. SVFC
It sounds as though they need to start another IND, specifically for MS. I am sure that those studies are very expensive. But if they might actually offer those suffering from MS, a great deal of relief, if not an actual cure, perhaps they should ask the MS foundation or some other foundation or government entity for a grant to do the study. The same goes for the brain injury patients, I would think that the pentagon might fund that study. I know that there are some bad things/hurdles for SVFC to overcome, but there is also so much in the pipeline that could accomplish such great things and help so many people that are suffering so badly. In my opinion, the good that could come of this, far outweighs the negatives. That is just my opinion. SVFC.

If true, yes. I would think that Dr. Victor would rather sell to Roche, rather than let one of these companies like Iron Ridge or YA Global take possession of his patents. I had thought that this would be discussed more than it has since Kallas first mentioned it this morning. I just hope that if Roche does purchase SVFC that it would actually do something with the technology. I am sure that we have all heard stories of very promising technologies that get purchased by some corporate giant because it would be easier to buy it and make sure that it is filed away and never heard of again, instead of having to compete with it.
Well, this has been discussed here as a real possibility. We have a board member at SVFC who was in a very powerful position at Roche. I know that most here have said that they would strongly prefer a partnership instead of a purchase. At least Roche would have the money to pay off all the debts. Considering all the things that have happened since this was last discussed on this board, how do you all feel about this happening? I have always been more than a little suspicious about a big pharma kind of company purchasing SVFC and then doing nothing with the technology.

BBW, you have said in the past that this was always about IR trying to get the patent that SVFC has, and pretty much everyone agrees with that. So how can this be a clear win for IR? They have likely spent more in attorneys fees than they will be receiving from SVFC. I cannot see how this is a clear win for them.

Yeah, me too. Thank you for the reply UncleRico. I have been wondering why everyone is discussing everything other than actual news regarding the EU patent. When I first looked at it, it appeared to be negative news, but then I thought that I might be misunderstanding it and that it might not be bad. I was hoping that someone here who understands all that legalese might take a look at it and give an educated opinion.
There appears to be some new material on the european patent site. They were filed yesterday. I am not a lawyer and I really don't know how to interpret these things.

The problem I have with that theory, which I hope is true, is that there is a regular poster here on this board who is an auditor, and he/she has said that it takes a full month to get this report finished after the last one is/was put out. Based on that statement, the next report still has a ~ week to go before it can be released. I hope that there is BIG news, maybe something like a partnership with Chaum, international patent granted, multiple agreements in place with other clinics around the world, etcetera. As always, time will tell.
I would not doubt it one bit. I was just reading a list of Conspiracy theories" that proved to be true. And several were false flag attacks that were meant/intended to sway public opinion. And a police officer friend of mine sent me a video regarding the Sandy Hook shooting that raised quite a few, very good questions. Such as a supposed parent of a slain child coming out a door to make a statement to the press. He is shown smiling and laughing as he comes out of a building, and then suddenly becomes very tearful while speaking to the press. And then it showed that this man was/is an actual actor. Another thing was a woman who was complaining that a photo of her child (that was already online) was used to show a picture of one of the supposed victims. The problem was that her child was just fine and they didn't live anywhere near that school. But it was an adorable photo, one that would work really well to tug at people's heartstrings. So yeah, there are quite a few unanswered questions regarding Sandy Hook. And our own government has been known to do this sort of thing in the past! I am of the opinion that if our government says so something, heck, anything, it is probably a lie!

This would be a pretty slick move on the part of President Obama, giving a grant to Intellicell Biosciences to hopefully help wounded soldiers coming home from the wars. Especially since he has been getting a lot of flack concerning the pitiful care that those soldiers have been getting at the VA hospitals. It would be a slick PR move on his part. Heck, it would be good for our soldiers, good for Intellicell Biosciences and for the President. And since this is a type of stem-cell therapy that does not use any kind of fetal stem-cells, it would also avoid the stigma of that whole mess. It would be a win for everybody concerned. Does anybody here have any political connections with the DNC? SVFC

I hardly ever see a nine hour gap between posts on this board. Looking at the 30 day chart, it looks to me as though SVFC has been going up since around May 27. I hope we close out the week with a strong showing. That regenerative medicine meeting that is going on today and tomorrow should send a lot of traffic to our SVFC. GLTA.
