Amarantus BioScience Reports Publication of Independent Peer-Reviewed Data for Receptor and Secretion Pathways related to MANF

Amarantus BioScience, Inc., a biotechnology company discovering and developing treatments and diagnostics for diseases associated with protein misfolding and apoptosis centered around its patented therapeutic protein Mesencephalic Astrocyte-derived Neurotrophic Factor (MANF), announced that an independent peer-reviewed research paper entitled Mesencephalic astrocyte-derived neurotrophic factor (MANF) secretion and cell surface binding are modulated by KDEL receptors was published in the Journal of Biological Chemistry.
According to a release, the research paper concludes "These findings provide insight into the mechanisms of MANF neuroprotection, and may be applicable to understanding its functions in other secretory tissues."
In addition, the Company noted:
The endoplasmic reticulum (ER) is the part of the cell where unfolded proteins are properly folded and then exported into different parts of the cell, or secreted out of the cell, so that they can perform their normal biological functions. When the ER becomes stressed, protein folding is compromised, and cells can become dysfunctional. Apoptosis, also known as Programmed Cell Death, is one of the main side effects of protein misfolding due to ER stress, and several independent peer-reviewed research papers have demonstrated that MANF plays a critical role in reducing protein misfolding and apoptosis. MANF has been shown to be upregulated inside the cell, and subsequently secreted out of the cell, with data demonstrating that MANF ultimately reduces apoptosis in the cells in which it originates, the cells it interacts with once secreted out of the cell, as well as when manufactured MANF is administered to various parts of the body undergoing ER stress. Amarantus is seeking to administer manufactured MANF to areas in the body where ER-stress occurs due to injury or disease, giving the body additional quantities of MANF during times of stress in order to reduce apoptosis and improve cellular function. The Company believes that if cellular function can be improved, overall system recovery is likely in many therapeutic indications. ER-stress and protein misfolding appear to be a key component of Parkinson's disease biology. Amarantus' Chief Scientist originally discovered MANF, and the Company was awarded composition of matter patents on MANF in Europe in 2010 and in the United States in 2011. The Company also recently won a court challenge to its composition of matter patents in Europe. The Company also owns worldwide patent applications covering various methods of using MANF as it relates to neurological conditions.
In this research paper, the authors describe the process by which MANF's intracellular activity (autocrine) may be regulated through MANF binding to the canonical ER-specific retention receptors with the amino acid sequence 'KDEL', known as 'KDELRs' (KDEL Receptors). Researchers expressed, or blocked, a key amino acid sequence on the MANF molecule known as 'RTDL,' which is a highly-conserved amino acid sequence that was shown in this research paper to be required for retention of MANF in the ER, or for secretion of MANF out of the ER, with signal activation through the KDELRs. Further, the research paper goes on to suggest that certain KDELRs are resident on the cell surface membrane, and that MANF's extracellular activity (paracrine) is regulated, at least in part, via cell-surface KDELRs. Interestingly, the paper suggests that following ER stress, there is an increase in expression of KDELRs on the cell surface membrane, opening the possibility that MANF's paracrine activity may be more potent or broad in times of ER stress.
"This publication provides further insight into the biological profile of MANF and the proteins it interacts with in both an autocrine and paracrine fashion," said Gerald E. Mesencephalic-Astrocyte-derived Neurotrophic Factor (MANF) is believed to have broad potential because it is a naturally-occurring protein produced by the body for the purpose of reducing and preventing apoptosis (in response to injury or disease), via the unfolded protein response. By manufacturing MANF and administering it to the body, Amarantus is seeking to use a regenerative medicine approach to assist the body with higher quantities of MANF when needed. A shipping vessel owned by Panamerican Seaways with capacity for 1,500 passengers and 500 cars, will begin travelling in May between Cartagena de Indias in Colombia and Colon in Panama.
Operators calculate that between 800 and 1,000 people will be carried on the "Nissos Rodos" ferry per trip, with rates starting from $99 and going up to $508.

The Caribbean cities of Colon in Panama and Cartagena de Indias in Colombia will be connected as of 10 May 2012 by a new ferry service, which will cover the shipping route in about seven hours, said the general manager of the company Aventuras 2000, Augusto Terracina. "The intention is to be able to connect these two countries which have always had a good relationship in both the tourist and commercial sectors," said Terracina, as part of a press conference to announce the launch. Terracina said the ferry "Nissos Rodos" of the cruise line Panamerican Seaways has a capacity to carry approximately 1,500 people, but they will start running at about half capacity, and they will also be carrying shipping containers and automobiles. He said the intention of Adventura 2000, which manages the Colon 2000 port in the Caribbean city of the same name, is to open Panama to the tourism and commercial landscape of the second country with the highest number of annual tourists. "We will be able to bring many more Colombian tourists from other regions, hoping this will become a mass product which attract groups for conventions, trips and other activities in Panama," he said. He said the investment is more than $15 million dollars, including making necessary improvements to the Colon 2000 port, improvements to the ferry ship itself, as well as the promotional aspect to be held in Panama and in Colombia. The inaugural departure is on 10 May 2012 from the city of Colon, and the next day there will be a similar ceremony at the port of Cartagena. Trips are scheduled to depart from either of the two ports at 6:00 pm local (23.00 GMT) and will arrive at 7:00 am local time (12.00 GMT) the following day. The ferry will make six trips each week, with three departing from Colon and three from Cartagena. The ferry will depart on Monday, Thursday and Saturday from the Panamanian port, and on Sundays, Tuesdays and Fridays from the Colombian city.
The trip on the Greek ship "Nissos Rodos" will have a cost ranging from $99 dollars for a "delux seat" to $508 dollars for a "presidential suite." The ship will also have the capacity for 500 cars and 2,000 square feet for cargo.
Terracina said the ferry will be manned by a Greek crew, who will be responsible to manage the boat, as well as Colombians and Panamanians who will be responsible for the restaurant, shopping and entertainment.
The first ferry crossings between the ports of Colon and Cartagena to promote trade and tourism in both countries were recorded between 1994 and 1997, but did not have the desired success due to several factors. In that sense, Terracina said tourism in Panama at that time was still emerging, and in Colombian there was the latent political problem between the government of that country and the guerrillas. But now the conditions of both countries are different, especially in Panama, where the tourist and commercial activity is in one of his best. (Panama America)

Hopefully the governments of both Panama and Colombia will make it easy for people to bring their cars with them if they want to - so you can go to Colombian and drive around for a week or whatever, without having to rent a car.
