I need a stiff whiskey and a biotech tutor to walk me through your thoughtful post. my summation: we need home run results from DF not just toleration, and marked improvement versus other drugs on the market. They need to show that tumors are regressing and stable. Those are a lot of big asks/wishes in a highly, highly, highly competitive field, so let's hope jim bonheim sinks a half court no second 3 at JP Morgan. a foul shot, or shovel pass for a first down, or decent serve and volley isn't going to wow anyone...1 month will be time to prove it big. is the drug working now and saving lives? aspirin in huge doses can be tolerated, we need to wow it up.

a masters or phd thesis ? Sounds like you know a lot about bio-pharmacology. You posted a lot of interesting info. Do you feel that CTIX and K will beat out the others ? When do you think that K will be available for patients ?

i remembered something else. names of spore grant recipients start being publicized in january. remember the pfizer sutinitab/kevetrin upcoming human trial at beth israel.

yes leo knows so does dr menon but what's better is he's telling us whatg he knows now.

whoops there are three labs doing qpcr now at DF. i think the top guy is dr herbert. leo said dr shipiro is now asking for the tests so that's another extra good sign. he sees something in those mri.s and serum markers.

hard to come down today. going to be a auphoric week.

Great post Orion! And don't go back down the rabbit hole! I don't recall anyone bringing up the heat shock proteins before and HSP90 pathway to tumor growth. You should try sending an email to Leo he might be able to answers some questions here. But
2 Things
First, if we were in college together would you do my homework for me?

Second, I think TruthbyThought and Ardhee are the same person.

Rabbit holes are dark and deep. Stay in the light. I didn't read the post too well but saw the poetry near the end...well done! I love that song, too. There is an excellent version of this on U-Toob where you can see the former exquisite beauty of one Grace Slick. By God, she is stunning! And what a voice!

Fantastic post detailing why K has the potential to have such a broad therapeutic index, it is not a one trick pony in terms of what pathways and regulators it can impact to fight cancer. This older PR provides further insight on MOA and the multiple means by which K can produce efficacy against treatment resistant cancers:

"Kevetrin™ has potent anticancer activity in a wide range of tumor types by targeting histone deacetylase (HDAC).

Cellceutix consultant Dr. Ashok Kumor recently made the discovery during the Company's research to further define Kevetrin's Mechanism of Action (MOA). Prior investigation of the MOA has shown that Kevetrin™ activates both transcription-dependent and transcription-independent pathways to induce apoptosis in tumor cells through activation of p53, the "Guardian Angel of the Human Genome," and potentially the retinoblastoma protein, or Rb, pathway. Damaged or mutated p53 or Rb is exhibited in nearly 100 percent of all cancers, regardless of origin. The HDAC2 connection further delineates the MOA behind Kevetrin's powerful antitumor activity that has been demonstrated against multiple cancers.

Inhibition or downregulation of HDAC reduces tumor growth and survival through cell cycle arrest, blocking of angiogenesis, an increase of antigenicity of tumor cells and induction of apoptosis. Given the involvement of deacetylase in cancer, inhibition or downregulation of HDAC has been investigated as a promising anti-cancer target.

In the study, Kevetrin™ potently downregulated HDAC2 in many mutant and null p53 cancer cell lines. HDAC2 is deregulated in many cancers and is emerging as the main deacetylase involved in aberrant pathways of tumor cells in humans. Downregulation of HDAC2 by Kevetrin™ strongly induced apoptosis in different types of tumor cell lines.

"We believe we have identified the Mechanism of Action of Kevetrin which explains why Kevetrin™ is so effective in a broad spectrum of cancers," says Dr. Krishna Menon, Chief Scientific Officer at Cellceutix. "Downregulation of HDAC2 also explains why Kevetrin™ is so effective in drug resistant tumors. Furthermore, since Kevetrin™ acts in a non-genotoxic manner and induces potent antitumor activity, we feel that Kevetrin™ is a clear standout from all other anticancer drugs. In all my years as a cancer researcher, I haven't seen a drug act like Kevetrin™ which is targeting and reacting with virtually every type of cancer."


Also, I leave tonight with a few simple quotes about K from Dr. Menon that speak for themselves (and i have to repost this one):

"In all my years as a cancer researcher, I haven’t seen a drug act like Kevetrin™ which is targeting and reacting with virtually every type of cancer."

"In all my years as a researcher, I have never seen anything like Kevetrin™.”

Coming from Dr. Menon these words carry a great weight, and have big implications. Implications that we are seeing come alive now at Dana Farber. :-)
