Because it was derived from a base compound that eventually was named Raltegravir before its commercial name Isentress. The base compound was discovered in the mid-1990s and Raltegravir entered Phase-1 testing around 1999. So, Isentress as the final commercial outcome has never been tested on anything but humans, or so the argument might go.
