AP113 & Sprycel/Tasigna progression

Hi Don,

Essentially what I was thinking on 113. Harvey's probably assuming that if croz gets approved, since it's the only ALK, our focus on the mutants(as in 534) should facilitate fast track 2nd line treatement. If I remember correctly, he even alluded to the parallels between the program prospects of 534 and 113.

But if croz fails(even better for ariad), he could be thinking we then qualify for fast track 1st line due to potential to meet completely unmet need regarding those 45,000 Alk driven NSCLC victims. Am I missing anything or making any false assumptions here?

Regarding sprycel/tasigna progression, I don't remember hearing that, so thanks for pointing it out. Assuming they continue to take market share from glivec, 534 could find itself at 500ml sales pretty quickly, not to mention the possibility that trial results and clinical experience may lead to more of a 1st line adoption of 534 by clinicians ahead of the official guidelines.

Regarding such a quasi off label usage, do you (or does anyone else here)know how much latitude clinicians have regarding use of the official 2nd line drug first if trial results suggest as good or better performance than the current 1st lines in an overall disease population, but with the added benefit of much better performance in cases where 1st line drugs are (for whatever reasons)ineffective? In other words, if others show the kind of enthusiasm for 534 that Dr K. did, do they have the option to employ a prospective 2nd line like 534 initially? Would of course be very important. Your thoughts?

On another matter,right now I'm kicking myself for not doing my homework on IDIX earlier, as I watch it rocket. Unfortunately, I still don't feel I know enough to take a position. Technically, it really seems to be breaking out from the 4.80s resistence on relatively high volume.

Another "Oh Well". Hopefully, I'll be up to speed for any retests.
