CHIMERIC vs HUMANIZED vs FULLY-HUMAN

TECHNICALLY:
”A therapeutic mAb can be chimeric, humanized or fully human. A Chimeric mAb contains a mixture of both mouse and human sequences, usually a 30/70 split, respectively, where the mouse components are responsible for binding to the antigen and the human components are involved in inducing a therapeutic effect. Humanized mAbs contain over 90% human sequences, while Fully-Human mAbs contain 100% human sequences.”

BUT PRACTICALLY:

“Antibody engineering also allows the production of "humanized" antibodies, in which only the gene sequences coding for the CDRs of the murine antibody replace the CDR sequences in human antibody genes. The resultant antibody is entirely human except for these short sequences and has essentially the same biochemical characteristics as native human [fully human] antibodies, as confirmed in practice. These antibodies display serum half-lives virtually indistinguishable from native human antibodies (14-21 days).”
