Israel (who is strongly suspected of possessing nuclear warheads..or even atomic bombs) will no doubt use nukes on Iran...if they feel that Iran will use them first against Israel.
As a matter of fact I do think Israel will strike first (using conventional means)....and they do think Iran is aggressively pursuing their own nuclear weapon...but I digress.

Make no mistake about it, if both nations possess nuclear/atomic WMD's, then both Iran and Israel will not hesitate to use nukes if they have to
Ditto for Pakistan. They're more of a regional threat though, IMO, than Iran is (thanks to an ancient, on-going border dispute with another nuclear power India--and the fact Pakistan has already demonstrated they possess nuclear tipped missiles).
