Good questions Thales! Entdoc, Mojo, Cj and many others can certainly give a more thorough and detailed response than I can.

However if you look at many of the patents you will see that Bavi can carry a wide variety of payloads. The list of potential payloads is mind boggling. It includes radiation,commonly used anti-cancer and antiviral drugs and a wide range of toxic cancer or viral killers that are so toxic they could not possibly be given systemically(eg.even RICIN!).

We have seen in all 3 phase II cancer trials that Bavi is used in combination therapy. But the drugs it is used with all have a wide range of toxic side-effects(just ask any cancer patient!). If Bavi could carry a payload of just these drugs directly to the cancer site...it could potentially dramatically reduce the horrible side-effects of systemic delivery. It could carry radiation to the cancer site( as Cotara does) and reduce the negative side-effects of radiation therapy. It could deliver powerful anti-viral drugs only to the virally infected cells and viruses...hence reducibg their very difficult side-effects(current HCV treatments are so difficult many patients withdraw from treatment!).

And yes fully human Bavi might be more potent...but because it is fully human might produce fewer adverse side-effects than the partly mouse Mab being used now. Some of the side-effects of Bavi in humans may be from the human body reacting to these foriegn mouse parts of the current Bavi. Wouldn't it be great if a more powerful Bavi with fewer side-effects produced a 90% ORR with 50% CR's!(just hoping!) I'm hopeful the fully human Mab would be both more powerful AND safer than the current version. It may not turn out that way. I really think Duke/Chavi and the DTRA sponsored trials may be looking at this in regard to the anti-viral indications. Once they get the phase I clinical studies underway and the safety of the fully human and/or armed fully human Bavi are proven...then they can be advanced on both the viral and cancer sides. Then some of our questions will be answered.

But even if this is not the case...the current Bavi is producing some very encouraging results in combination with many drugs in many difficult cancer indications!
