An observation from AMLN IV board this morning[that set of boards
not working well for me today]:

On NPR this morning, I heard a story about use of leptin injections in overweight (don't know if they were obese) people. The leptin reportedly affected the person's brain to increase (or bring back to normal) the "executive function" as opposed to the emotional response they normally have to food.
Scientists have been able to map brain activity of people who have lost weight through exercise/diet; when then presented with food, in absence of leptin injections, their response to food was highly emotional and the "executive" function was decreased - they really really wanted to eat and couldn't be rational about deciding whether to or not. If given the leptin, their brain's response to the food stimulus was the same as people who had not recently lost weight: activity in the emotional part of the brain was not higher and the activity in the "executive" part of the brain was normal.
This fits the results announced by Amylin/Takeda today: that patients [who] switched to placebo after losing weight with the pramlintide/metreleptin regained all of their weight.
My understanding is that pramlintide/metreleptin is like leptin on steroids[IMO, a curious simile]. And, it sounds like something one must take the rest of one's life in order to not gain the weight back. A real maintenance drug.<<
