Trevi Therapeutics advances cough treatment pipeline, eyes key clinical milestones

Trevi Therapeutics CEO Jennifer Good joined Steve Darling from Proactive to discuss the company’s progress in advancing its cough treatment pipeline using nalbuphine, a mixed agonist-antagonist opioid. Originally developed for acute indications, nalbuphine is now being repurposed due to its dual mechanism of action, working both peripherally in the lungs and centrally in the brainstem, offering a novel approach to treating chronic cough disorders. Trevi initially explored multiple indications, including skin conditions caused by severe itching and neurological cough. However, compelling clinical data led the company to focus on cough-related indications. Good explained, “We thought our drug was different because not only does it work peripherally in the lung, it works centrally at the brain, which mediates coughing and breathing.” The company is now targeting three key areas: interstitial lung diseases (including idiopathic pulmonary fibrosis, or IPF), refractory chronic cough (RCC), and other neurological cough conditions. Following promising Phase 2a results in RCC—an area where many previous drug candidates have failed—Trevi is preparing for a Phase 2b dose-ranging study. If successful, only one pivotal study may be required before regulatory filing. Tamboran Resources boosts NT gas supply with Arafura deal

Tamboran Resources Ltd CEO Joel Riddle talked with Proactive about the company's newly signed letter of intent (LOI) with Arafura Resources Ltd to supply gas from the Beetaloo Basin. Riddle explained that both companies are based in the Northern Territory, and the agreement supports local strategic projects by providing 25 terajoules (TJs) per day to Arafura's critical minerals project. The agreement aligns with Tamboran’s strategy of prioritising Northern Territory gas use. “We’re a Northern Territory company. We’ve been focused on appraising the Beetaloo Basin now for all 12 years I’ve been in my job,” Riddle said, highlighting the company’s long-term commitment to the region. Riddle also pointed out the environmental edge of Beetaloo Basin gas, which has a CO2 content of about 3%, significantly lower than the 15–20% typically found in other Australian basins. He said supplying this low-carbon gas to the East Coast would help reduce emissions by displacing higher-carbon alternatives. Discussing supply capacity, Riddle compared the Beetaloo to the Marcellus Shale in the US, noting it could provide enough gas to support the East Coast for “hundreds of years”. Tamboran plans to bring 40 TJs per day online in the first half of next year under an existing 15-year agreement with the NT Government. The Arafura deal will add a further 25 TJs daily. Riddle said work is underway with pipeline partner APA to ensure infrastructure capacity. The company aims to finalise a binding gas sales agreement with Arafura soon. Race Oncology launches Phase I RC220 trial in Australia

Race Oncology CEO and MD Daniel Tillett talked with Proactive about the opening of patient enrollment at the first Australian clinical site for the RC220 Phase I solid tumour trial. Tillett confirmed that the trial is now underway, with additional sites expected to follow soon. The focus of this early-stage study is to determine a safe and effective combination dose of RC220 with the chemotherapy drug doxorubicin. According to Tillett, “our drug has the advantage… not only does it improve the anti-cancer treatment, but it protects the heart, at least in animals and cells to date.” He noted the choice of Southside Cancer Centre as the initial site, citing the faster startup process compared to public hospitals. Other sites, including Gosford and Wyong, have also received ethics approval and are expected to join the trial following regulatory clearance. Tillett discussed the use of a Bayesian trial design, which allows quicker progression and minimises patient exposure to suboptimal doses. While this approach can introduce uncertainty in timelines, it is expected to be more beneficial overall for patients and the company. Early preclinical data has shown that RC220 can reduce doxorubicin-related toxicity while enhancing cancer treatment outcomes, a combination Tillett described as unique among current therapies. Beyond the trial, Race Oncology is conducting separate preclinical studies to identify the most effective drug combinations using RC220.
