Lenzilumab is a drug that works by blocking the action of a protein called granulocyte-macrophage colony-stimulating factor (GM-CSF). GM-CSF is a molecule that stimulates the growth and activation of certain types of white blood cells, such as neutrophils, eosinophils, basophils, and monocytes1. These cells are involved in the immune system’s response to infections and inflammation.
However, sometimes GM-CSF can cause an excessive or abnormal immune response, leading to harmful effects on the body. For example, GM-CSF can trigger a cytokine storm, which is a condition where the immune system releases too many inflammatory molecules into the bloodstream, causing damage to various organs and tissues. This can happen in some patients with COVID-19, a viral infection caused by the novel coronavirus SARS-CoV-22.
By blocking GM-CSF, lenzilumab can reduce the inflammation and prevent or treat the cytokine storm in COVID-19 patients. Lenzilumab can also help improve the survival and recovery of these patients, as well as prevent or reduce the need for mechanical ventilation2.
Lenzilumab is also being tested for other conditions where GM-CSF plays a role, such as acute graft versus host disease (aGvHD) and cytokine release syndrome (CRS) associated with chimeric antigen receptor T cell (CAR-T) therapy. These are both serious complications that can occur after stem cell transplantation or gene therapy, where the donor or modified cells attack the recipient’s own cells3 .
Lenzilumab is a humanized monoclonal antibody, which means it is a laboratory-made protein that mimics the natural antibodies produced by the immune system. Lenzilumab specifically binds to GM-CSF and prevents it from interacting with its receptors on the surface of white blood cells. This way, lenzilumab can modulate the immune system and restore its balance31.
