Trevi Therapeutics advances cough treatment pipeline, eyes key clinical milestones

Trevi Therapeutics CEO Jennifer Good joined Steve Darling from Proactive to discuss the company’s progress in advancing its cough treatment pipeline using nalbuphine, a mixed agonist-antagonist opioid. Originally developed for acute indications, nalbuphine is now being repurposed due to its dual mechanism of action, working both peripherally in the lungs and centrally in the brainstem, offering a novel approach to treating chronic cough disorders. Trevi initially explored multiple indications, including skin conditions caused by severe itching and neurological cough. However, compelling clinical data led the company to focus on cough-related indications. Good explained, “We thought our drug was different because not only does it work peripherally in the lung, it works centrally at the brain, which mediates coughing and breathing.” The company is now targeting three key areas: interstitial lung diseases (including idiopathic pulmonary fibrosis, or IPF), refractory chronic cough (RCC), and other neurological cough conditions. Following promising Phase 2a results in RCC—an area where many previous drug candidates have failed—Trevi is preparing for a Phase 2b dose-ranging study. If successful, only one pivotal study may be required before regulatory filing. Hillcrest launches ZVS technology evaluation with global Tier 1 automotive supplier

The project’s initial phase will assess the potential benefits of Hillcrest’s Zero Voltage Switching (ZVS) technology in next-generation inverter applications for electric vehicles. If this phase is successful and both parties agree to proceed, subsequent phases will focus on the integration and validation of ZVS technology within the supplier’s inverter platform. Currie explained that Tier 1 automotive suppliers provide fully assembled components and systems to major automakers. This particular supplier, consistently ranked among the top 15 largest by revenue, is a global leader specializing in critical EV components like inverters. With a strong presence in the industry, the supplier plays a crucial role in advancing electric vehicle technology for manufacturers worldwide. The evaluation phase will involve a series of technical assessments aimed at eliminating switching losses and improving electromagnetic compatibility (EMC) performance. Oriole Resources wraps up pivotal 2024, targets major gold milestones in 2025

Oriole Resources PLC (AIM:ORR) CEO Martin Rosser and CFO Bob Smeeton take Proactive's Stephen Gunnion through the company’s latest full-year results and upcoming milestones across its key exploration projects. Rosser highlighted a pivotal year for the company’s Bibemi and Mbe gold projects in Cameroon. At Bibemi, a recently completed phase five drilling campaign is feeding into a revised Mineral Resource Estimate (MRE), expected this quarter. “We believe there's very good potential to not just increase the amount of gold, but also improve the category of the resource,” said Rosser. The company is also advancing an exploitation license application for the project. At Mbe, maiden drilling continues, following trenching results that indicated strong mineralisation. “We think Mbe South has significant potential to host a substantial gold deposit,” Rosser added. Smeeton detailed a £2 million reduction in operating loss and financial support from BCM International. “Since year-end, BCM have given us $1.1 million, largely replenishing CapEx,” Smeeton noted. He also confirmed progress on a joint venture agreement with Managem at the Senala project in Senegal. Rosser pointed to potential sector-wide revaluations driven by high gold prices and possible M&A activity. “Oriole’s shares are significantly undervalued,” he said. Nexus Minerals expands Payns Find gold target

Nexus Minerals Ltd managing director Andy Tudor talked with Proactive about the latest exploration success at the Payns Find gold target, part of the company's Wallbrook Gold Project in Western Australia. Tudor described the program as “exciting” following a third air core drilling campaign that confirmed high-grade intercepts and significantly expanded the mineralised footprint. He said, “All of the attributes for a big system are here,” highlighting consistent grades of up to 5.99g/t at the extremities of a 900m by 750m target zone. The air core results revealed both supergene and fresh rock mineralisation, with around 30% of the 163 holes showing elevated geochemistry in the final metre — a strong indicator of a large underlying system. “It’s that grade in the supergene and in the fresh rock that’s got us excited,” Tudor said. He also pointed to the presence of a full suite of alteration minerals — cerussite, rutile, tourmaline, and albite — typically associated with major gold systems in the region. With the air core phase now complete, Nexus is turning to reverse circulation (RC) drill planning. Tudor confirmed that air core drilling would continue regionally, while the company prepares for a comprehensive RC program at Payns Find to test depth extensions down to 120–150m.
